乳品中常见过敏原特征多肽的筛查及制备应用

β-乳球蛋白是乳品中主要的过敏原物质,乳品过敏主要见于婴幼儿,严重时可能出现生命危险,是影响人体健康安全的主要因素之一。利用蛋白质组学数据库UniProt、酶解模拟软件Skyline、试验验证,对过敏原β-乳球蛋白的特征多肽进行筛查,筛选出1条稳定性强、抗干扰性能优越的多肽IDALNENK,并制备合成了13C、15N标记的IDAL*NENK(13C6, 15N-Leu)作为β-乳球蛋白的内标物。采用同位素稀释质谱法检测乳品中过敏原时将上述特征多肽作为内标物,可以准确定量乳品中β-乳球蛋白的含量,且灵敏度较高。

Abstract:

β-Lactoglobulin is the main allergen in dairy products. Dairy allergy is mainly seen in infants and young children and in severe cases may be life-threatening as well as being one of the main factors affecting human health and safety. Using proteomic database Uniprot, enzymatic hydrolysis simulation software Skyline and experimental verification, the characteristic polypeptides of the allergen β-lactoglobulin are screened.A polypeptide IDALNENK with high stability and superior interference resistance is screened, and 13C, 15N-labeled IDAL*NENK (13C6, 15N-Leu) is prepared and synthesized as a internal standard substance polypeptide for β-lactoglobulin. When using isotope dilution mass spectrometry to detect allergens in dairy products, the above characteristic polypeptide can be used as internal standard to accurately quantify β-lactoglobulin content in dairy products and with high sensitivity.
