提出了高效液相色谱法测定含氨基酸水溶肥料中游离氨基酸的含量。采用C18色谱柱(4.6 mm×250 mm,5 μm),以2, 4-二硝基氟苯为衍生剂,由水、乙腈和0.05 mol/L乙酸钠缓冲溶液(含体积分数1%的N, N-二甲基甲酰胺,pH=6.4)组成的流动相进行梯度洗脱,流量为1.2 mL/min,检测波长为360 nm。结果表明:17种氨基酸的质量浓度在1~200 μg/mL范围内与对应的峰面积呈线性相关,决定系数R2均大于0.996,检出限为1~3 mg/kg,加标回收率为83.3%~108.5%,相对标准偏差(n=6)为0.7%~4.5%。高效液相色谱法分离效果好、准确度高,可满足含氨基酸水溶肥料中游离氨基酸的检测要求。
A method for determining the content of free amino acids in water-soluble fertilizers containing amino acid using high performance liquid chromatography (HPLC) is proposed. The analysis is performed on a C18 column (4.6 mm × 250 mm, 5 μm) using 2, 4-dinitrofluorobenzene as the derivatization agent. Gradient elution is carried out with a mobile phase composed of water, acetonitrile, and 0.05 mol/L sodium acetate buffer solution (containing volume fraction 1% N, N-dimethylformamide, pH=6.4) at a flow rate of 1.2 mL/min. The detection wavelength is set at 360 nm. The results show that the mass concentrations of the 17 amino acids exhibited a linear relationship with their corresponding peak areas within the range of 1-200 μg/mL, with coefficients of determination (R2) all greater than 0.996. The detection limits ranged from 1 to 3 mg/kg, the spiked recovery rate are 83.3%-108.5%, and the relative standard deviations (RSD, n=6) are 0.7%-4.5%. The HPLC method demonstrates good separation efficiency and high accuracy, meeting the requirements for the determination of free amino acids in water-soluble fertilizers containing amino acid.
